Density Gradient Centrifugation Alone or the Combination of DGC with Annexin V Magnetic-Activated Cell Sorting Prior to Cryopreservation Enhances the Postthaw Quality of Sperm from Infertile Male Patients with Poor Sperm Quality

Objective. To examine whether density gradient centrifugation (DGC) alone or its combination with annexin V magnetic-activated cell sorting (DGC-MACS) can be used to process semen samples from infertile male patients poor sperm quality prior subjecting these freeze/thaw in order optimize the outcomes of freezing. Methods. This study enrolled sixteen concentration ≥ 20 × 10 6 / mL , id="M2"> motility < 30 %, and/or <4% normal morphology. Sperms were processed by DGC DGC-MACS process. Sperm motility, hyperosmotic swelling test (HOS), TUNEL test, and morphological analysis performed before after Results. The had a detrimental effect on viability, morphology, DNA integrity all three groups (RAW, DGC, + MACS groups). showed increased morphology following than untreated frozen controls. motility viability not significantly different between DGC-MACS-CPT (cryopreservation-thawing) DGC-CPT groups. Moreover, almost no grade A B was observed selected decreased levels fragmentation RAW freeze/thaw. group lower that group. Conclusions. preparation cryopreservation improved postthaw males quality. If is foreseen insufficient for artificial insemination husband’s vitro fertilization, if there high sperm, should reduce improve available intracytoplasmic injection.
